When conducting experiments using the Vole Sex Hormone ELISA Kit, it's important to pay attention to potential experimental errors. Here are the detailed steps recommended by Shanghai Jinma Technology to ensure accurate results:
1. Make sure all reagents are thoroughly mixed before use. Avoid creating excessive foam, as this can lead to air bubbles during sample loading, which might affect the accuracy of the results.
2. Determine the number of plates needed based on the number of samples and standards. It’s recommended to run multiple replicates for each standard and blank well. If possible, use duplicate wells for each sample to improve reliability.
3. Add 50 µL of the diluted standard and 50 µL of the sample into the reaction well. Then, immediately add 50 µL of biotinylated antibody. Cover the plate and gently mix to ensure even distribution. Incubate at 37°C for 1 hour.
4. After incubation, remove the liquid from each well and fill them with washing buffer. Shake for 30 seconds, then discard the buffer and pat dry with absorbent paper. Repeat this process three times. If using an automated washer, increase the number of washes by one.
5. Add 80 µL of streptavidin-HRP conjugate to each well, gently mix, and incubate at 37°C for 30 minutes.
6. Remove the liquid again, fill the wells with washing buffer, shake for 30 seconds, and discard the buffer. Pat dry and repeat the washing process three times. If using a washer, increase the number of washes by one.
7. Add 50 µL of both substrate A and B to each well. Gently mix and incubate at 37°C for 10 minutes. Be sure to avoid exposure to light during this step.
8. After incubation, quickly add 50 µL of stop solution to each well and measure the optical density (OD) immediately after adding the stop solution.
9. Finally, measure the OD value of each well at a wavelength of 450 nm to determine the concentration of the target hormone.
